| CM-Toyopearl陽イオン交換カラムクロマトグラフィーおよびSephacryl S-200ゲルろ過カラ ムクロマトグラフィーによって、常温性菌核病菌Sclerotinia sclerotiorumの20℃培養抽出物 のポリガラクツロナーゼ活性画分からポリガラクツロナーゼⅠを単離した。ポリガラクツ ロナーゼⅠ精製標品(P4-4-2)の比活性は3 662U/mgであり、粗抽出液から全体として116倍に 精製した。精製標品はポリアクリルアミドゲル電気泳動によって、均一であることが示さ れた。また、SDS存在下ポリアクリルアミドゲル電気泳動によって本酵素の分子質量は 39.8±1.10kDa(S.E.)、さらに等電点電気泳動によって本酵素の等電点はpH8.70と見積もられた。 Polygaracturonasehas been isolated from the mesophilic white mold Sclerotinia Sclerotiorum by cation-exchange chromatography on a column of CM-Toyopearl followed by gel filtration on a column of Sephacryl S-200.
